The development and optimization of efficient transformation protocols is essential in new citrus breeding programs, not only for rootstock, but also for scion improvement. Transgenic 'Hamlin' sweet orange (Citrus sinensis (L.) Osbeck) plants were obtained by Agrobacterium tumefaciens-mediated transformation of epicotyl segments collected from seedlings germinated in vitro. Factors influencing genetic transformation efficiency were evaluated including seedling incubation conditions, time of inoculation with Agrobacterium and co-culture conditions. Des observations et echantillonnages des hippocampes sont effectues dans le lagon du Grand Recif de Toliara, de l fannee 2006 a 2008, sur les trois especes d fhippocampes inventoriees : Hippocampus borboniensis, H. fuscus et H. histrix. Cette derniere espece est quasi absente dans la capture. H. borboniensis et H. fuscus sont recrutes respectivement a une longueur totale moyenne de 2,4 cm et de 3 cm correspondant a l fage de 3 et de 2 mois. La premiere maturite survient a une longueur totale moyenne de 6,1 cm a l fage de 8 mois pour H. borboniensis et de 5,58 cm a l fage de 8 mois et demi pour H. fuscus. The objective of this work was to characterize mandarin (Citrus spp.) germplasm from Southern Brazil by morphological and molecular analyses. Thirty seven cultivars from 34 distinct mandarin varieties were evaluated by morphological and agronomic traits of leaves, flowers and fruits, and by microsatellite markers. The morphological and agronomic characteristics suggested that almost all varieties can be produced for commercial use, and some, as the Satsuma variety, are recommended for breeding programs. The purpose of this work was to compare the SPD (Single Pod Descent) and SPDS (Single Pod Descent with Selection) methods for segregating generation advancement in soybean breeding. Sixteen populations derived from crosses among Japanese and adapted Brazilian soybean were used. The traits, number of days to flowering (NDF) and to maturity (NDM), plant height at flowering (PHF) and at maturity (PHM), and individual plant yield (IPY) were assessed. The populations obtained by SPDS showed values close to the variation in the populations obtained by SPD. Regression through the origin showed that both methods were equivalent for NDM in the F3 generation and for NDF in the F4 generation. A estimação de componentes de variância e a predição de valores genéticos no contexto do melhoramento de plantas perenes, demanda o uso da metodologia de mode los mistos (REML/BLUP) ao nível individual, a qual vem sendo aplicada com eficiência ao melhoramento de várias espécies perenes. Uma questão fundamental na aplicação dessa metodologia refere-se à definição dos efeitos do modelo estatístico como fixos ou aleatórios. No caso do efeito de repetições experimentais esta definiçâo nem sempre é trivial. Assim, esta questão é abordada no presente artigo.

O sucesso dos programas de melhoramento genético depende dos procedimentos de seleção e dos métodos de melhoramento adotados para a condução das populações segregantes. O objetivo deste trabalho foi avaliar a eficiência do método Bulk com seleção em F3, (BulkF3) em relação ao método Bulk, bem como comparar qual estratégia de seleção é mais eficiente em termos de ganho genético. Random amplified polymorphic DNA markers (RAPD) were used to estimate the variability of 14 genotypes of Brazilian wheat (Triticum aestivum L.), using a set of 50 random 10mer primers. A total of 256 reproducibly scorable DNA amplification products were obtained from 48 of the primers, 83% of which were polymorphic. Genetic distances among genotypes were calculated and a dendrogram and a principal coordinates analysis showing the genetic relationships among them were obtained. Despite the low variability found (average genetic distance of 27%), two groups of genotypes could be identified, which probably reflect how they were formed. The objective of this work was to construct linkage maps of 'Pêra' sweet orange [Citrus sinensis (L.) Osbeck] and 'Cravo' mandarin (Citrus reticulata Blanco) using RAPD markers and the pseudo-testcross strategy. The parents were chosen according to the resistance/susceptibility to citrus variegate chlorosis (CVC). The segregation of 176 markers was analyzed in 94 progeny of F1 hybrids, which were obtained from controlled crossings. The linkage map of 'Pêra' sweet orange had 117 markers defined by 12 linkage groups, which spanned 612.1 cM. Only six markers could not be linked to the linkage group and 48.7% of the markers showed segregation distortion. The linkage map of 'Cravo' mandarin had 51 markers defined by 12 linkage groups, which spanned 353.3 cM....

It is common sense, however, that this progress can be improved further if refined breeding techniques, developed from the knowledge of the genetic mechanisms controlling soybean yield, are used.
